What's your system specs, by the way? Please post your graphics card too. It all depends on your video card(vc). Your vc will give you information about the resolutions that it can display on your monitor. You can easily do this via:
Control Panel --> Display --> Settings --> and look at the "Screen Resolution" lever. Try to drag it and see which resolutions it can support. The color settings would also tell you if you can display at 16-bit or 32-bit color resolutions.
With a low-end vc, you won't notice anything when you change to 16-bit resolution, except that the gameplay performance might improve a little. With a high-end (or a mid-end) vc and a good monitor such as Mag Innovision or ViewSonic, you can notice an amount of detail loss especially at higher screen resolutions (1024x768 or beyond). High-end vc's might not even give you the option to display lower color settings such as 16-bit. If you can't change the 32-bit in-game resolution, then perhaps you have a vc that can support higher screen resolutions and color settings. The vc driver might find it insignificant to give you lower options since your system can afford to display graphics at high resolutions.

I agree with curious. There's no real gain or loss by switching it to 16bit.
