I don't know anything about boxout strength so I won't be talking about it.
About the defensive assist, from my knowledge I know that the setting you use in controller settings reflects into the online game too. In my opinion, when setting the defensive assist near 100, the game helps you stay in front of your opponent (when you press LT) when he does basic sprinting around and stuff. So basically, you don't have to move the left stick for every step, sometimes your player moves by its own in the best defening position. From what I've noticed though, if your opponent starts doing iso moves, the chances are he will run right past you. The defensive assist for iso moves is underpowered, you fell like you become the AI on rookie difficulty; you go in the opposite direction when he crosses, you freeze when he does spinning moves.
Summary - Big defensive assistive strength -> Good positioning against basic sprinting, bad for ISO moves. That's why 2K has no problem in connecting opponents with different assist settings; it's a pick your poision type of situation.
So the best idea would be of course to set the assist to 0 and try to stay in front of your opponent by your own, using a combination of LT, the left and the right stick, and sometimes the sprint button. You move your you want to, so you can't blame the game for your mistakes.
Personally, I have the defensive assist on 50 during online games because of the lag. When a patch will be released and online games will run smoothly, I will probably set it to 0 as I do in offline games.
